They probed the sample with an innovative experimental technique that revealed the three-dimensional architecture of the mammoth\u2019s genome. The&nbsp;<a href=\"https:\/\/www.cell.com\/cell\/fulltext\/S0092-8674(24)00642-1\" rel=\"noreferrer noopener\" target=\"_blank\">resulting paper<\/a>&nbsp;was published on Thursday in the journal Cell.<\/p>\n\n\n\n<p>Hendrik Poinar, an evolutionary geneticist at McMaster University in Canada, was \u201cfloored\u201d \u2014 the technique had successfully captured the original geometry of long stretches of DNA, a feat never before accomplished with an ancient DNA sample. \u201cIt\u2019s absolutely beautiful,\u201d said Dr. Poinar, who reviewed the paper for the journal.<\/p>\n\n\n\n<p>The typical method for extracting ancient DNA from fossils, Dr. Poinar said, is still \u201ckind of cave man.\u201d It produces short fragments of code composed of a four-letter molecular alphabet: A (adenine), G (guanine), C (cytosine), T (thymine). An organism\u2019s full genome resides in cell nuclei, in long, unfragmented DNA strands called chromosomes. And, vitally, the genome is three-dimensional; as it dynamically folds with fractal complexity, its&nbsp;<a href=\"https:\/\/www.scientificamerican.com\/article\/untangling-the-formation-of-dna-loops\/\" rel=\"noreferrer noopener\" target=\"_blank\">looping<\/a>&nbsp;points of contact help dictate gene activity.<\/p>\n\n\n\n<p>\u201cTo have the actual architectural structure of the genome, which suggests gene expression patterns, that\u2019s a whole other level,\u201d Dr. Poinar said.<\/p>\n\n\n\n<p><a href=\"https:\/\/www.nytimes.com\/2024\/07\/11\/science\/mammoth-dna-genetics.html\">Read it all<\/a>.<\/p><blockquote class=\"twitter-tweet\"><p lang=\"en\" dir=\"ltr\">My latest for <a href=\"https:\/\/twitter.com\/Nature?ref_src=twsrc%5Etfw\">@Nature<\/a>: On Siberia&#39;s frozen tundra some 50,000 years ago, a woolly mammoth met its end.
